This project represents a follow-up to Pathos-17, using microbiological, immunological, and transcriptomic approaches (Dual-RNASeq) on peripheral and intestinal dendritic cells (DCs), infected and uninfected by various bacterial mutants, derived from healthy individuals and Crohn’s disease patients. Aims include: 1) understanding microbiological, immunological, and molecular pathways, which regulate the persistence, in peripheral/intestinal DCs, of E. coli AIEC with the consequent activation of pathogenic Th1/17 cells; 2) characterizing the immune responses of E. coli AIEC-infected DCs in healthy individuals and Crohn’s patients; 3) functional validation of RNA targets, which promote AIEC-DC interaction/Th1/17 activation in the intestine; 4) formulation of preclinical models for the development of next-generation immunotherapies for Crohn’s disease.
