High-Grade Serous Ovarian Cancer (HGSOC) is the most frequent among ovarian cancers. Ribosomal protein (RP) L8 coding gene (RPL8) is frequently amplified (27%) and correlates with miRNA-6850 (included in an intronic portion of the RPL8 gene) amplification. RPL8 is a structural component of ribosomes, whose qualitative and quantitative alterations have been linked to cancer, besides to ribosome-independent functions. Our preliminary data indicate that copy number alteration of the RPL8 gene may play a role in HGSOC. In addition, miRNA-6850-5p is highly expressed in HGSOC, but not in other OC types, and its levels in serum have been correlated to a poorer prognosis. The main objective of this proposal is to unravel the molecular mechanisms that link RPL8 genetic amplification to OC, be they linked to RPL8 ribosomal or extra-ribosomal functions, or rather to miRNA-6850, or to a combination of these mechanisms.
